Our vision is health independence and wellbeing for every person on the planet with neurological conditions including Autism Spectrum Disorder diagnosis. BRAIN Foundation UPCOMING Synchrony 2025 Premier annual conference on translational research in autism Autism Spectrum Disorder ASD is the fastest growing developmental disability 1 in 36 children is diagnosed with autism in the US and larger numbers have speech and language impairments learning difficulties and other related impairments. Yet there are only two FDA approved treatments for autism both addressing only a narrow scope of surface symptoms.

Prenatal infections have long been recognized as risk factors for #autism. Infectious agents such as rubella, HIV, cytomegalovirus and Zika virus may disrupt neurodevelopment through inflammation, brain #metabolism and #epigenetics changes. Epidemiological studies show that maternal infections during pregnancy are linked to higher autism risk, regardless of the infectious agent involved, with the strongest association in cases requiring hospitalisation.

“Genes set the stage and the environment pulls the trigger” is likely true for many diseases and disorders, including #autism. The two-hit, or multiple-hit theory of autism would also go a long way towards explaining the heterogeneity of the disorder and the presence (or absence) of co-occurring conditions in autism, such as intellectual disability, seizures and others. A good example of this is Tuberous sclerosis complex (TSC), a syndrome caused by inactivating mutations in the genes encoding TSC1/2, negative regulators of the mammalian target of rapamycin complex 1 (mTORC1).
